Dysregulation of the TGF-β Pathway in Pancreatic Cancer and Colorectal Carcinoma: A Comparative Analysis

Abstrakt/ Abstract:
Cieľ/ Objective: The aim of this article is to analyze the mechanisms of TGF-β signaling pathway dysregulation in two serious types of cancer, PDAC and CRC. Although both types of cancer are among the most common and deadly in the world and share an epithelial origin, their molecular characteristics differ. Material and methods: The study used the results of gene expression of selected members of the TGF-β pathway in tissue homogenates obtained from the Gepia gene database.
Výsledky/ Results: Gene expression analysis in PDAC has revealed extensive and consistent downregulation of key components of the pathway, including ligands (TGFB1, TGFB2, TGFB3), receptors (TGFBR1, TGFBR2), and intracellular signaling proteins (SMAD2, SMAD3). Unlike PDAC, CRC exhibits a different pattern of dysregulation. The expression of many components (TGFB1, TGFB2, TGFBR1, TGFBR2, SMAD2, SMAD3, SMAD4) remains unchanged. However, there is significant upregulation of the TGFB3 ligand, TGFBR3 receptor, and negative regulator SMAD7.
Záver/ Conclusion: These changes suggest that the dysregulation of TGF-β signaling in CRC is not about an overall weakening of the pathway, but rather a selective shift that can lead from tumor-suppressor function to pro-tumorigenic function. These findings point to the need for a specific approach to targeted therapies.
